Emergency services were called to the Hume Freeway near Harrys Creek Rd following a report a B-double truck was involved in a collision just before 1.30pm.
Police believe the truck collided with a road worker’s truck and then veered across the median strip where it clipped a car, before it rolled and caught fire.
The driver of the B-double truck, who is yet to be formally identified, died at the scene.
The driver of the second truck, and the occupants of the car, were transported to hospital with non-life-threatening injuries.
Ambulance Victoria has confirmed two adults and two children were taken to Northeast Health in Wangaratta by road ambulance.
A man in his 50s was taken by road ambulance to Goulburn Valley Health in Shepparton with minor injuries.
All five were in a stable condition when transported to hospital.
The exact circumstances surrounding the collision are yet to be established, and an investigation is ongoing.
