The biennial event is normally held on even years, however after COVID-19 stopped last year's crawl taking place organisers decided to break from tradition.
That decision was assisted by the organisers of Hauling the Hume, the event that normally happens on odd years travelling the opposite direction, cancelling their event.
The crawl sets off from the old Ford factory in Campbellfield around 6.15 am and follows the route of the old Hume Highway up to Albury.
Trucks have to be at least 25 years of age to participate in the event.
Event organiser Robert French said technically this was still the 2020 Crawling the Hume, with registrations for the event closing 14 months ago.
"Last year we were in the shed two weeks before the crawl packing everything up ready to go when the phone started to go off," Mr French said.
"I thought we'd be right ... but then we got news that gatherings were restricted to less than 200 people and we had to pull the pin on it.
"We always planned to go ahead with the event, but for a long time we didn't know when."
All trucks need to be on the road before 8 am, with the slower trucks setting off first.
They will be going through Seymour mid-morning then up to Benalla, stopping at Winton Motor Raceway for lunch.
Truck fans are encouraged to visit Winton to see what is on show and have a chat to drivers.
Mr French said after lunch the trucks would set off en-route to Albury where participants would enjoy a dinner and speeches from a number of guest speakers.
"On Sunday morning drivers will attend a breakfast and awards ceremony with tongue-in-cheek awards handed out, including slowest driver, worst gear change," he said.
"Then we all say goodbye and trucks will head off in every direction."
